BDNF基因多态性与冠心病共病抑郁的关联及其血小板活性机制

摘要:

目的:探讨BDNF基因rs6265基因多态性与冠心病共病抑郁的关联,及其血小板活性机制。方法:采用HAMD-17对冠心病患者共病抑郁情绪进行筛查,收集到冠心病共病抑郁患者49例和冠心病未共病抑郁患者124例,收集患者临床资料,检测患者血小板活性,抽取患者外周静脉血,提取DNA,采用TaqMan探针SNP基因分型技术,对BDNF基因rs6265的单核苷酸多态性进行检测。结果:BDNF基因rs6265位点基因型分布频率,和等位基因频率在冠心病共病抑郁组与冠心病未共病抑郁组之间有显著差异,冠心病共病抑郁组突变基因型AA分布频率和突变等位基因A频率显著高于冠心病未共病抑郁组;且共病抑郁可使冠心病患者血小板比积更高。是否携带A等位基因对血小板活性的影响主效应均无统计学意义。结论:中国冠心病患者中携带BDNF rs6265等位基因A者对共病抑郁更易感,但本研究未发现其通过作用于血小板活性而参与共病机制。

Objective:To investigate whether polymorphisms of BDNF influence depressive mood and platelet activity in patients with coronary artery disease(CAD). Methods:Clinical data of 49 CAD patients with depressive mood and 124 CAD patients without depressive mood were collected. Peripheral blood samples were drawn and their DNAs were collect-ed. Rs6265 single nucleotide polymorphisms(SNPs) of BDNF gene were detected through TaqMan SNP genotyping assay in all subjects. Results:Significant difference in genotypic and allelic distribution of rs6265 were found between CAD patient with depressive mood and those without depressive mood, BDNF rs6265 AA genotypic distribution and allelic A distribu-tion were significantly higher in CAD patient with depressive mood group than those without depressive mood group. And comorbidity with depressive mood made CAD patient higher platelet pressure. No significant main effect of genotype was found on platelet activity. Conclusion:CAD patients with A allele in BDNF rs6265 single nucleotide polymorphisms are more vulnerable to depressive mood, but not found through the platelet activity pathway in this research.
